Minor Groove Binder (MGB) probes are superior and specific alternatives to SG probes (SYBR Green). The major disadvantages of intercalating probes such as SG are that they bind to all double stranded (ds) DNA and are non-specific in detecting primer dimers and other contaminants. High concentrations of SG also results in inhibition of PCR reactions. SG or other DNA intercalating probes prove to be useful only in the analysis of whole DNA, due to lack of specificity.
tinto rang™ is a minor groove / external binding nucleic acid dye that has a high afficnity and rapidly binds to DNA. It can find potential applications in the making of safe, fluorescence-based MGB probes that have several advantages over DNA intercalating probes in terms of increased stability, sensitivity and specificity.
